Designing guides one target at a time is tedious, and it's tricky to get it right: you have to check GC content, avoid poly-T runs, screen for cross-reactivity between targets, and separately design genotyping primers for each one. This is a batch tool that does all in one pass.Point it at a FASTA file (or a folder of them) and get back ranked gRNA candidates, off-target checks against your other targets, and ready-to-order validation primers, for as many genes as you want, in one run.What it does: Finds every candidate guide matching your PAM (SpCas9 NGG by default; supports NG, SaCas9, and Cas12a) Scores candidates on GC content, poly-T avoidance, hairpin risk, and seed-region quality; tells you what happened in the output Checks for cross-reactivity between your target sequences Auto-designs validation primers (via primer3) flanking each cut site Processes any number of targets in one command How: There's no lightweight open-source version of Doench 2016ML model that Benchling/CRISPOR use; so this pipeline uses a documented heuristic scoring model. You can also find full explanation of scoring and off-target scope in the README. It's a good tool to batch-screen and shortlist; you shouldnt use this to replace experimental validation.Includes a README, example data, and --help flag.
